Falmouth RFC will face fellow Western Counties West club Saltash in the first round of the 2019/20 Tribute Cornwall RFU Knockout Cup.
The tie will be played at the Rec on or before November 3.
Penryn are one of four clubs that have been handed a bye to the quarter-finals, alongside St Austell, St Ives and defending champions Wadebridge Camels.
Wadebridge Camels won the Tribute Cornwall Cup for the first time in seven years when they defeated holders St Ives 30-26 in April.
